概要

Shikon root extracts have been used to treat chaps, heat rash, incision and burns. Migration, proliferation and differentiation of keratinocytes are important processes during wound healing of the skin. Our aim for this study was to determine effect of shikonin on the gene expression profile during artificial wound healing of keratinocyte HaCat. Cultured in confluence HaCat cells were scratched by plastic comb in the presence or absence of shikonin, and were harvested 3 hours or 16 hours following injury. RNA was extracted and reverse transcribed into cDNA and subject to cDNA microarray analysis of 54,359 cDNA probes. At 3hours, in the presence of shikonin resulted in consist increase (> 2 fold) in gene expression of 409 (0.75 %) or that of decrease (< 0.5 fold) in gene expression of 1,261 (2.3 %) out of 54,359. At16 hours, artificial wound was almost repaired (> 95% area) and 1,242 cDNAs were increase (> 2 fold) and 1,065 cDNA were decreased (< 0.5 fold) by treatment of shikonin. Three hundred and three genes were down regulated at 3 hours and upregulated at 16 hr by shikonin. Five genes were constitutively down regulated and 40 genes were up regulated at both period. This is the first report that shikonin selectively regulates gene expression during wound healing process. Relationship between shikonin treatment and expression profiles of wound related gene will be discussed.
